Protoporphyrinogen oxidase (PPO)-inhibiting herbicides remain an important and useful chemistry 60 yr after their first introduction. In this review, based on topics introduced at the Weed Science Society of America 2021 symposium titled “A History, Overview, and Plan of Action on PPO Inhibiting Herbicides,” we discuss the current state of PPO-inhibiting herbicides. Renewed interest in the PPO-inhibiting herbicides in recent years, due to increased use and increased cases of resistance, has led to refinements in knowledge regarding the mechanism of action of PPO inhibitors. Herein we discuss the importance of the two isoforms of PPO in plants, compile a current knowledge of target-site resistance mechanisms, examine non–target site resistance cases, and review crop selectivity mechanisms. Consistent and reproducible greenhouse screening and target-site mutation assays are necessary to effectively study and compare PPO-inhibitor resistance cases. To this end, we cover best practices in screening to accurately identify resistance ratios and properly interpret common screens for point mutations. The future of effective and sustainable PPO-inhibitor use relies on development of new chemistries that maintain activity on resistant biotypes and the promotion of responsible stewardship of PPO inhibitors both new and old. We present the biorational design of the new PPO inhibitor trifludimoxazin to highlight the future of PPO-inhibitor development and discuss the elements of sustainable weed control programs using PPO inhibitors, as well as how responsible stewardship can be incentivized. The sustained use of PPO inhibitors in future agriculture relies on the effective and timely communication from mode of action and resistance research to agronomists, Extension workers, and farmers.

Background
Studying phenotypic and genetic characteristics of age at onset (AAO) and polarity at onset (PAO) in bipolar disorder can provide new insights into disease pathology and facilitate the development of screening tools.
AimsTo examine the genetic architecture of AAO and PAO and their association with bipolar disorder disease characteristics.
MethodGenome-wide association studies (GWASs) and polygenic score (PGS) analyses of AAO (n = 12 977) and PAO (n = 6773) were conducted in patients with bipolar disorder from 34 cohorts and a replication sample (n = 2237). The association of onset with disease characteristics was investigated in two of these cohorts.
ResultsEarlier AAO was associated with a higher probability of psychotic symptoms, suicidality, lower educational attainment, not living together and fewer episodes. Depressive onset correlated with suicidality and manic onset correlated with delusions and manic episodes. Systematic differences in AAO between cohorts and continents of origin were observed. This was also reflected in single-nucleotide variant-based heritability estimates, with higher heritabilities for stricter onset definitions. Increased PGS for autism spectrum disorder (β = −0.34 years, s.e. = 0.08), major depression (β = −0.34 years, s.e. = 0.08), schizophrenia (β = −0.39 years, s.e. = 0.08), and educational attainment (β = −0.31 years, s.e. = 0.08) were associated with an earlier AAO. The AAO GWAS identified one significant locus, but this finding did not replicate. Neither GWAS nor PGS analyses yielded significant associations with PAO.
ConclusionsAAO and PAO are associated with indicators of bipolar disorder severity. Individuals with an earlier onset show an increased polygenic liability for a broad spectrum of psychiatric traits. Systematic differences in AAO across cohorts, continents and phenotype definitions introduce significant heterogeneity, affecting analyses.

Much of the solid waste stream in the United States is generated by metropolitan areas, while associated landfills are often located in adjacent rural communities. Landfill disposal of municipal solid waste often creates external costs to nearby residents. Contingent valuation was used to estimate external costs of siting a landfill in the Carter community of Knox County, Tennessee. Estimates of annual external costs were $227 per household. Household income, size, years in the community, and distance from the proposed landfill and the respondent's education, sex, and perception of health risks were important in determining a household's willingness to pay to avoid having a landfill in the Carter community. Also, households whose (drinking water supplies were at risk of contamination were willing to pay $141 more than those who used piped city water or bottled water.

A cement-based grout (90% Type 50, 10% silica fume, 0.4 < water-to-cement ratio, w/c < 0.6) has been used in field trials at AECL's Underground Research Laboratory in Canada and at the OECD/NEA Stripa Mine in Sweden, to evaluate suitable grouts and grouting techniques that could be used for sealing a nuclear fuel waste disposal vault mined deep in granite. Laboratory studies have been carried out to determine the following grout properties: hydraulic conductivity (k); resistance to piping and erosion during setting; influence of grout on the pH and chemical composition of water permeating grouted rock; and the ability of the grout to self-seal after fracturing.
Laboratory tests have confirmed the low intrinsic k of these cement mixtures (10−14 m/s). Using a specially developed cone-in-cone apparatus, we have studied the effect of fracture dilation and temperature changes on the k of thin films of cement. If fractured, the grout has an ability to self-seal and the rate of self-sealing increases with increasing temperature.
The pH and ionic composition of the water permeating grouted fractured granite rock were found to vary with grouted fracture aperture and grout/rock volume ratio. The field tests demonstrated that grout can penetrate and seal very fine fissures (apertures less than 50 μm).
